Guarnaschelli was the daughter of cookbook editor Maria and John Guarnaschelli. She graduated from Barnard College in 1991 with a degree in art history. For one year, she was paid minimum salary at an American Place in 1991. Guarnaschelli began her career in the culinary field by helping her mother to test many recipes and then writing cookbooks. Guarnaschelli's career began in the kitchen of Larry Forgione. Her son Marc Forgione was the Iron Chef. Guarnaschelli then went on to work at various restaurants throughout France as well as Los Angeles. Guy Savoy runs La Butte Chaillot. She also worked at Daniel Boulud's own restaurant, as well as Joachim Splichal's Patina prior to becoming Chef-in-Charge at Butter. After the restaurant's closure, she was the executive chef of The Darby. She is chair of the Museum of Food and Drink's Culinary Council. Guarnaschelli published her first cookbook in the year 2013. "Old-School Comfort Food": The Way I learned to cook" blends personal details and some of her favorite dishes from her professional life that she made for the family home.
